Transmitted strains may have evolutionary advantage, NIH scientists suggest

WHAT:

A new study has found that even though HIV diversifies widely within infected individuals over time, the virus strains that ultimately are passed on through heterosexual transmission often resemble the strain of virus that originally infected the transmitting partner. Learning the characteristics of these preferentially transmitted HIV strains may help advance HIV prevention efforts, particularly with regard to an HIV vaccine, according to the scientists who conducted the study. The research was led by Andrew D. Redd, Ph.D., staff scientist, and Thomas C. Quinn, M.D., senior investigator, both in the Laboratory of Immunoregulation of the National Institute of Allergy and Infectious Diseases (NIAID), part of the National Institutes of Health.

Dr. Redd and colleagues examined particular genetic sequences of HIV in blood samples collected between 1994 and 2002 from hundreds of HIV-infected heterosexuals participating in the Rakai Community Cohort Study in Rakai District, Uganda. The scientists found a significant increase in population-wide HIV genetic diversity among infected individuals during the 8-year period, yet the amount of genetic change was significantly greater within individuals than across the population as a whole. To explain this phenomenon, the scientists hypothesized that the genetic diversity of HIV at the population level was limited because only certain strains of the virus within each person were responsible for subsequent sexual transmissions.

To test this hypothesis, the scientists examined the genetic relatedness of HIV strains in 31 couples where heterosexual transmission occurred. Through three increasingly comprehensive analyses, they compared HIV strains in the transmitting partner at points before and around the estimated time of HIV transmission with the strain in the newly infected partner around the time of transmission. In 22 of the couples, or 71 percent of those studied, the virus strains in the blood of the newly infected partner were more closely related to those found in the blood of the transmitting partner at the earliest available time point than to strains present around the time of transmission. According to Dr. Redd, this finding demonstrates that in the heterosexual transmission of HIV, the frequent natural selection of viral strains from early in the infection of the transmitting partner reduces viral diversity at the population level.

Moreover, in four couples, the newly acquired strain was highly similar or identical to specific variants found in the transmitting partner at both the earliest time point and the time of transmission. The scientists hypothesize that these highly transmissible HIV strains from early infection were sustained in the blood at low levels or sequestered in certain cells for transmission at a later time.

Related research by other scientists shows that HIV strains found in infected individuals during the early stages of infection have diversified little from the strain that caused infection. Thus, the fact that these early HIV strains somehow are maintained or persist at low levels for transmission later suggests they may have an evolutionary advantage at crossing the genital barrier and causing infection, compared with HIV strains that predominate later in infection, according to Dr. Redd.

This research was co-funded by the NIAID Division of Intramural Research; NIAID grants numbers R01A134826, R01A134265 and R01AI077473; the World Bank STI Project; the Henry M. Jackson Foundation; the Fogarty Foundation; and the Bill and Melinda Gates Institute for Population and Reproductive Health.

Priorities in couple relationships

by Jonathan on 09/18/2012

If you were to identify your top ten priorities in your life in order of importance, what would those be? Or, if you are filling this out as it relates to your spouse, what do you honestly believe they would list as their top ten? You could include things such as work, children, spouse, church, volunteer work, hobbies, self-care, use of social media, time spent with friends, watching television, caring for family members, school, and many others.

Now, imagine I am a fly on the wall that could observe your day to day activities, or the activities of your spouse. ?If I did this for two weeks I would get a fairly accurate feel for how priorities are handled on a daily basis. ?What would I list as the real top ten priorities based solely on my observations of your behavior or the behavior of your spouse? Be honest and list what you think my observations would be. ?It can also be helpful for you and your spouse to complete this list about each other as well

Are the lists the same? I would be surprised if they are. ?Most have good intentions and priorities are simply not reflected in the reality of stressful day to day life. ?There are just not enough hours in the day. ?However, I suggest that the behaviors most accurately reflect your properties. ?The behaviors are the priorities you decided to actually make a priority and put to action.

I try to respect that everyone is unique and has different needs. ?Assuming that to be true, one would suggest that the best priority order cannot be easily generalized. ?If the goal is simply to respect individuality, then the argument may be that there should not be a best way to prioritize. ?However, in the interest of maintaining healthy relationships, I believe there is a best way to prioritize. ?The simplest way to do this is to always put your spouse as your number one priority. ?Period.

This does not mean that your spouse gets more of your time than anything else, because work demands, parenting responsibilities, and other things may often take more of your time. ?However, research has consistently shown that people who make their spouses their number one priority are much less likely to have marital relationship problems. ?This then has a ripple effect on all involved, including children.

China surveillance ships near islands disputed with Japan

BEIJING/TOKYO (Reuters) ? Six Chinese surveillance ships briefly entered waters near disputed islands claimed by Tokyo and Beijing on Friday, raising tensions between Asia?s two biggest economies to their highest level since 2010.

Japan protested to China and urged that the situation not be allowed to escalate ? an outcome neither side would welcome given the two countries? tight economic links.

Diplomats say Tokyo and Beijing would prefer to keep the row from spiraling out of control, but with China facing a once-in-a-decade leadership change, an election looming in Japan and mutual mistrust deep, managing the feud could be difficult.

?The dangers of miscalculation are real,? said Brad Glosserman, executive director at Honolulu?s Pacific Forum CSIS.

China?s foreign ministry said that the ships entered the disputed waters to conduct maritime surveillance and that for the first time China was carrying out a mission of ?law enforcement over its maritime rights?.

The Japanese coast guard said it ordered the Chinese ships to leave the area. By afternoon, all had left the area without any use of force, a coast guard official said.

The islands, known as Senkaku in Japanese and Diaoyu in Chinese, are near potentially huge maritime gas and oil fields.

?We lodged a strong protest and also we made a strong case that the Chinese side should leave from the territorial waters around the Senkaku islands,? Japanese Foreign Minister Koichiro Gemba told a news conference in Sydney after talks with Australia?s foreign and defence ministers.

?I?d like to underscore that we should never let the situation escalate and we have strong hopes for the Chinese to respond in an appropriate and calm manner,? he added.

Chinese ambassador Chen Yonghua, summoned to Japan?s foreign ministry to hear a protest, repeated Beijing?s stance on the islands but added it also hoped the situation would not escalate or hurt ties, a Japanese foreign ministry statement said.

The uninhabited islets were at the centre of a chill between Beijing and Tokyo in 2010, after Japan arrested a Chinese trawler captain whose boat collided with Japanese Coast Guard vessels in the area.

Sino-Japanese relations have long been plagued by China?s bitter memories of Japan?s military aggression in the 1930s and 1940s and present rivalry over resources and regional clout.

TENSION THREATENS TRADE

China warned Japan on Thursday that trade could be hurt by the flare-up. China, the world?s second-largest economy, is Japan?s biggest trading partner with mutual trade in 2011 growing 14.3 percent in value to a record $345 billion.

Tensions have risen since nationalist Tokyo Governor Shintaro Ishihara floated a plan for metropolitan authorities to buy the islets and build facilities on them. That prompted Prime Minister Yoshihiko Noda?s government to buy them instead.

Japan deported Chinese activists who landed on the isles last month, triggering a tit-for-tat landing by Japanese nationalists and anti-Japanese protests in Chinese cities.

On Tuesday, Japan, which controls the islands, finalised their purchase from a private owner, ignoring warnings from China that the move would breach its sovereignty.

Japan?s consulate in Shanghai said on its website at least four Japanese citizens had been injured in attacks stemming from the tensions and warned Japanese in the city to be careful.

Small protests continued on Friday in front of the Japanese embassy in Beijing, with groups of about 40 people shouting anti-Japanese slogans and waving Chinese flags.

Cheng Li, an expert on Chinese politics at the Brookings Institution think tank in Washington, expressed concern about the rise in tensions.

?We cannot eliminate the possibility of military conflict,? Li said. ?Chinese leaders actually talk tough and act carefully, but sometimes it?s out of your control. Chinese public opinion has become so powerful.?

Washington has expressed concern, this week urging both sides to tone down their increasingly impassioned exchanges. (Additional reporting by Ben Blanchard in Beijing, Kiyoshi Takenaka in Tokyo and Damian Gill in Sydney; Editing by Jeremy Laurence)

Twitter hands over Occupy Wall Street protestor's updates under pressure

Twitter has been building a modest reputation for siding with the little guy (or girl) when it comes to communication privacy, and it just demonstrated how far it's willing to go in a showdown with Manhattan's Criminal Court over a demand to hand over tweets from Occupy Wall Street protestor Malcolm Harris. The social network has been pushed into delivering the claimed evidence, but only as it faced a deadline and the threat of a fine -- it even tried one last request for a stay before producing hard copies of the messages. However much the handover affects Harris' chances at winning during trial, it emphasizes that public posts have serious consequences -- companies ultimately can't shield you from the law.
